What Is Syn-Ake? Benefits, Research & Safety

Syn-Ake was developed by Pentapharm, a Swiss biotechnology company specialising in active ingredients for cosmetics and pharmaceuticals. The development was inspired by research into the Temple Viper (Tropidolaemus wagleri) and its unique venom composition. The Temple Viper, native to Southeast Asia, produces a venom containing waglerin peptides that cause reversible muscular paralysis. Unlike most snake venoms that work through enzymatic destruction of tissues, waglerin causes a non-destructive, reversible blockade of nicotinic acetylcholine receptors—making it scientifically interesting for cosmetic applications. Pentapharm's researchers sought to create a synthetic peptide that could replicate waglerin's receptor-blocking activity without the full complexity or potential risks of the natural venom peptide. The result was Syn-Ake—a small tripeptide that competitively binds to nicotinic receptors. The peptide was introduced to the cosmetic market in the mid-2000s and quickly gained attention for its novel mechanism. Unlike Argireline and other SNARE-targeting peptides, Syn-Ake offered a completely different approach to neuromuscular modulation. Pentapharm was acquired by DSM (Dutch State Mines), a global science-based company, which continues to manufacture and market Syn-Ake as part of their cosmetic ingredients portfolio. The peptide has been incorporated into numerous premium anti-ageing products worldwide. The "snake venom" marketing angle has made Syn-Ake popular in consumer products, though scientists emphasise that the synthetic peptide shares only the mechanism—not the risks—of actual venom components.
