The kisspeptin story began in 1996 when the KISS1 gene was identified as a metastasis suppressor gene in melanoma cells—hence the original name "metastin." The gene was found at the Hershey Medical Center in Pennsylvania, leading to the playful name "KISS1" (a reference to Hershey's Kisses chocolates). For several years, KISS1/metastin was studied primarily in cancer research. The discovery of its critical role in reproduction came in 2003 when two independent groups discovered that mutations in the KISS1 receptor (GPR54) caused hypogonadotropic hypogonadism—failure to undergo puberty. This discovery fundamentally changed understanding of reproductive neuroendocrinology. Kisspeptin was rapidly established as the key upstream regulator of GnRH release, effectively serving as the "on switch" for puberty and the gatekeeper of reproductive function. Subsequent research revealed that kisspeptin neurons integrate multiple signals—including metabolic status, stress, and day length—to appropriately regulate reproduction. This helps explain why conditions like anorexia nervosa or extreme exercise suppress menstruation. Active clinical research is ongoing into kisspeptin's therapeutic potential, with studies investigating its use in IVF, hypothalamic amenorrhoea, and as a diagnostic tool for reproductive disorders.
